IRF1-AS overexpression inhibits PPRV replication. A, C EECs and E, F GFFs were transfected with pCDNA3.1 plasmids or pCDNA3.1-IRF1-AS plasmids for 24 h and then infected with PPRV at an MOI of 3 for the indicated times. The RNA expression levels of IRF1-AS (A, E) and the protein levels PPRV N expression (C, F) were measured by quantitative RT-PCR and Western blot assay, respectively. B EECs were transfected with increasing amounts of pCDNA3.1-IRF1-AS plasmids (0, 2, 4 µg) for 24 h, and the cells were infected by PPRV (MOI = 3) for 48 h. The protein levels of PPRV N were measured by Western blot assay. D EECs were transfected with pCDNA3.1 plasmids or pCDNA3.1-IRF1-AS plasmids for 24 h and then infected with PPRV at an MOI of 3 for the indicated times. At the indicated time, the virus titres in the supernatants were measured by TCID50 assay. β-actin was used as a loading control in Western blot analysis. The data represent the mean ± SD of three independent experiments. P values were calculated using Student’s t test.
