IRF1-AS downregulation enhances PPRV replication. A, B EECs and E, F GFFs were transfected with si-NC or si-IRF1-AS-1 and si-IRF1-AS-2 siRNA for 24 h and then infected with PPRV at an MOI of 3 for 48 h. The RNA expression levels of IRF1-AS (A, E) and the protein levels PPRV N expression (B, F) were measured by quantitative RT-PCR and Western blot assay, respectively. C EECs and G GFFs were transfected with si-NC or si-IRF1-AS-1 for 24 h and then infected with PPRV at an MOI of 3 for the indicated times. The protein levels PPRV N expression were measured by Western blot assay. D EECs were transfected with si-NC or si-IRF1-AS-1 for 24 h and then infected with PPRV at an MOI of 3 for the indicated times. In the indicated time, the virus titres in the supernatants were measured by TCID50 assay. β-actin was used as a loading control in Western blot analysis. The data represent the mean ± SD of three independent experiments. P values were calculated using Student’s t test.
