From: ASFV pD345L protein negatively regulates NF-κB signalling by inhibiting IKK kinase activity

pD345L suppresses the mRNA expression of IFNβ and proinflammatory cytokines. A Quantitative real-time PCR was performed with RNA extracted from 293 T cells transfected with pcDNA3.1-HA-cGAS and pcDNA3-2xFlag-STING along with p3XFLAG-CMV-D345L or empty vector for 24 h. B WSL or C PK15 cells were transfected with p3XFLAG-CMV-D345L or empty vector for 30 h followed by treatment with 2′3′-cGAMP (5 μg/mL) for 4 h and then harvested for RNA extraction. Semiquantitative RT–PCR assays were carried out to detect IL-1α, IL-6, IL-8, IFNβ, TNFα, D345L and Actin mRNA expression levels. The levels of IFNβ and proinflammatory cytokines were normalized to the level of actin.
