From: African swine fever virus MGF360-11L negatively regulates cGAS-STING-mediated inhibition of type I interferon production

ASFV MGF360-11L could inhibit the IFN-β signaling pathway. cGAS (500 ng) and STING (500 ng) plasmids were co-transfected into HEK-293 T cells (A) and PK-15 cells (B), along with MGF360-11L (1 μg) or empty vector plasmids. Twenty-four hours post-transfection, IFN-β, ISG15, and ISG56 mRNA was analyzed by RT–PCR. C MGF360-11L (1 μg) or empty vector plasmids were co-transfected into PK-15 cells for 18 h, and then the cells were infected with HSV (MOI = 1) for another 8 h. RT–PCR was conducted with the indicated primers. D MGF360-11L (1 μg) or empty vector plasmids were co-transfected into 3D4/21 cells, after 24 h post-transfection cells were stimulated with 2′3′-cGAMP for 12 h, RT–PCR was conducted with the indicated primers. All experiments were independently repeated at least 3 times. The data are shown as the mean ± SD; n = 3. *p < 0.05, **p < 0.01, ***p < 0.001.
