Flowchart of the experimental setup used to sort Tetracapsuloides bryosalmonae from infected kidney of fish and RNA sequencing. Brown trout and rainbow trout were experimentally challenged with laboratory cultured T. bryosalmonae. Ten weeks post-exposure kidney samples were collected and cell suspension was made from them. Cell suspension from kidney was stained with anti-Tetracapsuloides bryosalmonae antibody and anti-mouse antibody Alexa 488 and then subjected to sorting by FACS Aria III cell sorter. Samples were then used for RNA extraction and cDNA library preparation followed by sequencing. Subsequently, data was analyzed using bioinformatics approaches to identify differentially expressed genes of T. bryosalmonae between the two fish species.
